Abstract

Bir ilaç etkin maddesinin, kozmetik ürünün, çevresel bir kimyasalın (pestisitler, endokrin bozucular, ağır metaller, nanopartiküller gibi), ağır metallerin ve fiziksel veya biyolojik ajanların in vitro olarak sitotoksik etkilerinin güncel yöntemlerle belirlenmesi son yıllarda oldukça önem kazanmıştır. Sitotoksisitenin veya hücre canlılığının belirlenmesi için birçok yöntem bulunmaktadır. Bu yöntemler (i) boyama yöntemleri; (ii) kolorimetrik yöntemler; (iii) florometrik yöntemler; (iv) luminometrik yöntemler; (v) apoptozun belirlenmesi için kullanılan farklı teknikler ve (vi) otofajinin belirlenmesi için kullanılan farklı teknikler olarak sınıflandırılabilir. Bu yöntemlerin hangisinin en uygun yöntem olduğu araştırıcı tarafından tüm bilimsel veriler kullanılarak değerlendirilmelidir. Bu değerlendirme yapılırken test maddesinin fiziksel ve kimyasal özellikleri, maddenin hangi mekanizma ile hücre ölümüne yol açtığı, yönteminin özgünlüğü ve hassasiyeti dikkate alınmalıdır. Bu şekilde elde edilen sonucun güvenirliliği ve doğruluğu konusunda emin olunmalıdır.
