Alzheimer Hastalığında Antosiyaninlerin Önemi

Abstract

Giriş: Yirmi birinci yüzyılın en önemli sağlık sorunlarından biri haline gelen Alzheimer demansında ana patalojik belirteçler amiloid plaklar ve nörofibriler yumaklardır. Beyinde belirgin atrofi mevcuttur. Ayrıca ek olarak inflamasyon, oksidatif stres, apoptoz, mitokondri hasarı, bozulan kalsiyum homeostazisi de süreçte rol oynamaktadır. Günümüzde Alzheimer hastalığının tam sağaltımını sağlayacak bir tedavi yöntemi henüz bulunmamıştır. Alzheimer hastalığının çok faktörlü patogenezi göz önüne alındığında, tek hedefli tedavi yaklaşımları yetersiz kalmaktadır. Etkin bir tedavi için hastalığın ilerlemesine katkıda bulunan birden fazla faktörü hedefleyen tedavi yaklaşımlarının benimsenmesi gerekecektir. Galantamin, Rivastigmin gibi kolinesteraz inhibitörü olarak etki gösteren lisanslı ilaçların çoğunun, fitokimyasallardan alkoloid bazlı ilaçlar olması, bitki kaynaklı ilaçlar konusunda kapsamlı araştırmalar yapılabilmesi için emsal teşkil etmektedir. Ayrıca,kolinerjik yetersizliği düzeltmenin ötesinde oksidatif stres ve nöroinflamasyonu da kontrol altına almak demans insidansını azaltmanın anahtarlarından biri olabilecektir. Bitkiler tarafından üretilen antioksidan ve antiinflamatuar özellikte olan sekonder metabolitlerin bu noktada önemli koruyucu etkileri olduğu bildirilmiştir. Polifenoller, bitkiler tarafından sentezlenen sekonder metabolitlerdir. Antosiyaninler; bitkilerde en önemli flavonoid grubu içerisinde yer alan pigmentlerdir. Antosiyaninlerin, oksidatif ve nitrosatif stres, eksitotoksisite, glial inflamasyon, protein agregasyonu ve apoptotik sinyal proteinlerinin indüksiyonu gibi nörodejenerasyonda yer alan süreçlerin zararlı etkilerinin çoğunu iyileştirdiğini gösteren çeşitli bulgular ortaya çıkmıştır.
Sonuç: Antosiyaninler hem diyetsel bir müdahale hem de gelecekteki ilaç keşif çabalarının temeli olarak terapötik potansiyele sahip olabilir ve bu da onları nörodejenerasyon için tedaviler geliştirme bağlamında dikkate alınması gereken ilginç bir bileşik haline getirmektedir.
